However, if are not already using an approved eManifest Service Provider you will have to register with them first.

No. CBSA requires all drivers to wait one full hour after the ACI eManifest is accepted before approaching the border. Failure to wait one hour could result in AMPS penalties from CBSA. eManifest App has a countdown clock for all accepted eManifests to ensure that our customers are aware of how much time they have remaining before they can cross.
